Есть, рассматривается как категория 3 согласно GAMP5: Другой вопрос, что лично я не сталкивался с ситуациями именно по 1С, чтобы они в фарма не нуждались как минимум в кастомизации (а это уже категория 4), а чаще - полноценная доработка функционала по некоторым специфическим фарма-процессам (тогда это категория 5). Но, тут зависит, действительно какой "коробочный" продукт вы берёте. Те же склады вы ведь настраиваете под себя - вряд ли они в коробке есть именно ваши. Но последняя фраза - это наугад, конечно. Так же это могут быть, например, бизнес-процессы по смене статусов "карантин"/"брак"/"разрешено" по сырью и готовой продукции или же по заполнению аналитических листов (сертификации номенклатуры). Я не видел таких "коробок" 1С, не нуждающихся в допиливании, во всяком случае, пока )). Не забываем, что интересна не столько их коробочность, сколько их GxP-релевантность. Например, закрытие неоперативного проведения для GxP-транзакций - ведь если эту возможность не закрыть, то это будет "выстрел в висок" по целостности данных. Разграничение прав доступа? Ведь это тоже не в коробке. Аудиторский след тоже, скорее всего, будет нуждаться в доработке, поскольку в полной мере родной контрольный журнал не будет соответствовать требованиям Приложения 11 GMP (указание причины при изменении GxP-релевантных данных - те же данные по статусам и/или аналитическим листам).Вот с этим тезисом я был бы поосторожнее с любой системой - потому что следующий вопрос - может ли поставщик предоставить по запросу отчет о тестировании такой любой системы? 🙂 Например, производители CDS (хроматографических систем данных) - могут. Поставщики решений типа Elpro, Vaisala, Testo Saveris Pharma - могут. От 1С отродясь ничего подобного не видел. Но, возможно, что-то поменялось в этом отношении или именно вашем случае это иначе. Но в любом случае - тесты от поставщика - это хорошо. Но "жить" ведь будет 1С на вашей ИТ-инфраструктуре? Если да, то как минимум стандартные, "коробочные" GxP-релевантные бизнес-процессы нужно воспроизвести у вас под конкретными пользователями, документы и справочники тоже будут вашими - это и будет основной объем валидации, если всё-таки категория 3 по GAMP 5. Ну и квалификация вашей ИТ-инфраструктуры. | |||||

@hendler, в ГОСТ 56893-2016 (перевод ISO 17665-2) указано, что как правило реквалификация осуществляется ежегодно (там термин переаттестация, но не суть - это "вечный привет переводчикам" - переводы одного и того же пункта 12.4 ISO 17765-1 и ISO 17665-2 выполнены по разному, но первоисточник и смысл один и тот же - requalification): Можете в вашем случае постараться обосновать другой интервал, но это будет источником постоянного регуляторного риска - для одной инспекции ваше обосновании будет достаточным, а для другой, возможно, нет. | |||||

PDA TR No. 1 - стерилизация влажным жаром, PDA TR No. 3 - стерилизация сухим жаром/депирогенизация. Вот выкопировка из PDA TR No. 3 в части базовой температуры стерилизации сухим жаром: При этом дополнительно если брать ЕР и ГФ РБ (упомянув ТКП, мы ведь к РБ адресуемся, верно?), то в статье (5.1.1) по стерилизации, в пункте по сухожаровой стерилизации указана базовая температура именно 160 С и время - 2 часа. А вот в части депирогенизации согласно PDA TR No. 3 базовая температура уже 250 С и время - 30 мин, что должно обеспечивать снижение бакэндотоксинов на 3 порядка. Поэтому тут есть определенный теоретический диссонанс - ЕР (и ГФ РБ) говорят о температуре более 220С, а PDA TR No. 3 - о температуре 250 С. Но формула Fd (F-value for depyrogenation) есть только в PDA: Формула для Fd (по тексту PDA, правда, пишут именно как F-value for deyrogenation - но суть та же) такая же, как и для Fh, только с базовой температурой 250 С и z = 46,4C. Подставив в формулу вы получите Fd = 30 мин. Если начнете повышать температуру - то получите время, необходимое для соответствующей температуры, например, для 300 С Fd ≈2,5 минуты. P.S.: ТКП, как правило, являются компиляцией в т.ч. указанных технических отчетов PDA - рекомендую именно документы PDA использовать в качестве первоисточника. ТКП удобны и практичны, но, к сожалению, не лишены опечаток (например, ТКП 433 в части базовых температур стерилизации/депирогенизации и соответствующих им величин z). | |||||

F0 - это всегда привязка к базовой температуре 121 С (ну, строго говоря, 121.1 С), может вы имеете в виду Ft - время при конкретной температуре, когда через скорость летальности можно сопоставить с эквивалентным воздействием при температуре 121С? Условно: Ft = F0/L - выше в комментариях есть пример, что если мы используем стандартный режим ЕР (121С-15 минут), то для достижения такого же эффекта при 110С понадобится 193 минуты с копейками: | |||||

Традиционно хронически не хватает времени, вместе с тем пока полностью (без редактирования и сокращений) выложу мою переписку с членами TC 209 WG 3. На этот раз Mr. Gordon Farquharson переадресовал наше обращение своим коллегам и ответ был получен от Mr. Stephen Ward, технического директора британской компании Validair. Вот его ответы по тексту нашего исходного письма (в соавторстве с Михаилом Шаховым): Hello Oleksandr, Thank you for your comments. As an update, the ISO14644-3 DIS, has been revised since the issue you have commented on. There is actually a 2nd DIS being prepared for issue (it is currently in its translation process). Although your comments may not have been addressed in the 2nd DIS, it would be worth you reading this once it is issued and you can comment formally and this will allow the working group to consider and review your comments. I am not sure how you officially direct comments in Russia to ensure they are in your countries formal comments back to ISO. It needs to be directed this way to ensure they can be included and formally reviewed. Russia does have a representation in the working group. Either professor Fujji or perhaps Gordon Farquharson can give you the contact information if they have it. I have however made a few comments which are just my opinions and may not reflect those of ISO the working group or the UK. See my comments below. Dear Mr. Gordon Farquharson, first of all let us introduce ourselves - this letter is initiated by Oleksandr Belinskyi (ISPE Member 374040 since 2015), Head of validation department LLC “LabPromEngineering” (Minsk, Belarus) and Mikhail Shakhov, specialist for qualification, NPC “Cleanroom Instruments” (authorized dealer of Lighthouse Worldwide Solutions in Russia) Our letter is intended to clarify the procedure described in clause B.7.3 Procedure for installed filter system leakage scan test with a DPC (ISO/DIS 14644-3 Cleanrooms and associated controlled environments -- Part 3: Test methods) and, probably to add some more practical information concerning this test. Primary it worth say a few words concerning the benefits of suggested revised procedure: ISO/DIS now allows clearly distinguishing Ca and Car values in clause B.7.3.1 ISO/DIS determine Cc value expressed in particles/m3 as it is in mostly commercially available DPCs The scan rate (Sr) is formulated as “5 cm/sec or less” without complex calculations, that actually are senseless for manual maintaining of this value ISO/DIS determine Ca using 0,3 µm particles size But we would like also suggest a few comments for given procedure: ISO/DIS contains in clause B.7.1.1 the common link to IEST-RP-CC034.4 - it’s useful link. But the mentioned recommended practice concerning this procedure in clause H1 DPC IN-SITU FILTER SCAN TEST METHOD has the following note: “This test is typically limited to cleanrooms where the actual air cleanliness classification is ISO Class 5 or cleaner. Testing the perimeter of filters in less clean areas may result in false indications of leaks due to the induction of particles from the high ambient particle concentration. It may be possible to use a barrier to shield the filter perimeter from the aerosol contribution from the room.” The ISO/DIS doesn’t consider this note - which “barrier to shield” you can suggest? Could it be at least the construction similar to airflow hood for balometers or something like that: How to perform the scanning using such “barrier to shield”- it’s another consequent question. The main idea is to prevent false indications that you could hardly distinguish in a case of small leak detected in ISO 8 surrounding. (SCW: I agree, there can be a problem of false counting when testing with an LSAPC (DPC) in lower class areas. I do not think the Working Group has thought to suggest barrier methods and perhaps this is best left to the individual decide the most effective and appropriate method based on the individual situation) We suggest not to choose primary Ca and corresponding Np in order to calculate Cc but to do just on the contrary - first measure Cc (knowing that the desire order is about 108 particles/m3) - and then calculate the Ca and Np: Why so? The reason that the upstream concentration is not easy to adjust and maintain according to the definite calculated value and excessive upstream concentration can cause false leakage detection. (SCW: I think in reality, this is what does happen, but this way makes the formulae understanding a little more logical) The most important part of our letter. We suggest significant addition for procedure simplification from practical point of view. Current standard version and ISO/DIS contain procedure B.7.4 (B.6.4 in the current ISO 14644-3:2005) Procedure for overall leak test of filters mounted in ducts or air-handling units (AHUs). In spite of the ducts dedication there is the following foreword present: “These tests may also be used for terminally mounted filters as long as they are positioned in installations with a non-unidirectional flow regime”. (this statement has been removed from the 2nd DIS) Also the clause B.7.4 contains the note: “It is important to be aware that this procedure is far less sensitive at finding leaks than the method described in B.7.2 and B.7.3”. Theoretically we agree, especially when the overall leakage doesn’t allow to determine definite leakage location. But practically only small leakage has additional risk to be not detected at all. We have done (LLC “LabPromEngineering”) the series of test for comparison of methods B.7.3. and B.7.4 for the same HEPA-filters of the same HVAC (the same AHU). We have used the above mentioned airflow hood not for scanning but for integral measurement - it also worth to mention this in B.7.4 because ISO/DIS allows this method for terminally mounted filters but doesn’t describe how to perform this definitely for terminally mounted filters. The result of both methods were closely correlated. The benefit of the B.7.4 - you obtain the estimation much more quickly, the surrounding room doesn’t has an influence in a case of ISO Class 7 or 8 and if even integral evaluation (overall leakage method) results the the filter system has a leakage - than you switch to scanning and for sure find the source. This hint could be useful for practical implementation. (I can see a benefit, but as we have now removed the earlier statement, we would not add this) ISO/DIS costitues that the method B.7.4 is far less sensitive, but using EN 1822-1 criteria integral leakage criteria is five time more stringent than the local one: But we would like to go furthermore and suggest the “mixed” practical method - scanning, with optionally used “barrier to shield” in a case of ISO Class higher than 5, but estimating downstream concentration (not absolute counting using relatively complex calculations) directly. We measure upstream concentration, than calculate the desired acceptance criteria using given above table from EN 1822-1 using simple proportion: for scanning we use local penetration value for overall leakage we use integral penetration value That’s all. This ISO/DIS suggest for overall leakage estimation in clause B.7.4 (but not describes it in details - it worth to add). When scanning we suggest to add additional simplified procedure, for example, as clause B.7.5 (before Apparatus and materials). Example: we have H14 HEPA filters. Using Table 1 from EN 1822-1 we can obtain it’s integral effectiveness as 99.995 %, and it’s local effectiveness as 99.975 %. So “allowed” leakage during scanning should not exceed 0.025 %. Having Cc = 1.8*108 particles/m3 we obtain: 1.8*108 * 0.025 % = 4500 particles/m3. Simple. Practically achievable and, the main aspect, the sensitivity is the same when you perform scanning with absolute counting and more complex preliminary calculation. Note: we understand that EN 1822 give us the effectiveness based on MPPS, that normally less than 0.3 µm, but the current methods also consider this situation acceptable for in-situ measurements. Additional benefit when using directly downstream concentration evaluation - you can adjust on your DPC continuous sampling mode instead of 1 sec probe sampling duration. Actually this situation also needs to be clarified - when you obtain your Ts value about 0.2 or 0.4 sec - how do you estimate your Ca? The reason is that most of DPCs has minimum sampling time 1 sec - does it means, that we need to multiply Ca by 5 or by 2.5? And we need to perform the scanning in intermittent mode - 1 sec stay then scan, 1 sec stay then scan? Otherwise all our theoretical formulations are not exactly correct in spite of still good possibility of leakage determination. The minor but also important aspect. When you perform leak test using DPC you normally take to account cumulative values, but not the differential values. Why it is so - for cleanrooms monitoring the personnel used to do so, for recovery test - also. Everywhere cumulative values are considered. When you perform leak test it’s not correct. Example. Imagine you have 1 million 0.3 µm particles and 1 million 0.5 µm particles before filter. Let’s assume that we have 50 % effectiveness for 0.3 µm particles and 100 % 0.5 µm particles (for simplify the calculations only - the real ratio is for sure differs from this assuming). In this case we obtain after the filter 500 thousand particles (all 0.3 µm due to 50 % effectiveness, 0.5 µm will be absent totaly because of 100 % effectiveness). Using cumulative values from DPC we obtain 75 % effectiveness (from overall initial 2 millions particles we obtain after the filter only 500 thousands), but using differential value our effectiveness will give us only 50 %. So it worth to mention that the point of interest differential values (mode) on DPCs. And the latest but not the last. Short consideration concerning procedure with aerosol photometer usage - why the standard suggests 0,01 % as acceptable leakage for any applications? The matter is we can test H13 filters, H14 filters, U15 filters - the filter efficiency is different according to EN 1822, but the acceptance criteria for mass concentration is always the same. (SCW: A complicated subject and one that could result in pages of response. It is contentious, but consideration is being given to the fact that this is a test to find leaks caused by damage or poor installation and not to verify that a U16 is performing to U16 levels. When you have this philosophy in your head, it’s a case of what level of leak compromises my cleanroom and perhaps a standard 0.01% across the board addresses that approach better than, “I have installed a U16 so I need to test to U16”.) Возник вопрос по валидации конфигурации 1С. Если мы покупаем "коробочный" продукт и собираемся использовать его в том виде в котором он поставляется (на 100% соответствует URS без изменения конфигурации), есть ли необходимость в его валидации? Ведь по факту он уже должен быть многократно протестирован службой качества поставщика ПО. Может быть достаточно в этом случае провести IQ + проверку СОП + проверку резервного копирования? Если же поставщик "дописывает" под нас какие-то процессы, которые являются GxP-критичными - то проводим интеграционное тестирован ... | |||||

... протестировать код и предоставить результаты тестирования вам. Вы, по сути, тестируете (валидируете) правильность работы ПО по функциям и процессам. Для этого у вас есть URS и FS. |
